Living Biointerfaces for the Maintenance of Mesenchymal Stem Cell Phenotypes

Living interfaces are established as a novel class of active materials that aim to provide an alternative traditional static cell culture methods by enabling users accurately control behaviour in precise, dynamic, and reliable system-internal manner. To this day, the only reported biointerface has been coculture between biofilm nonpathogenic genetically engineered bacteria mammalian cells, where recombinant proteins produced directly influence behaviour. In work, is presented Lactococcus lactis (L. lactis) human mesenchymal stem cells (hMSCs). L. have produce C-X-C motif chemokine ligand 12, thrombopoietin, vascular adhesion protein 1, 7th–10th type III domains fibronectin, with recreating native bone marrow conditions ex vivo. This microenvironment shown maintain key hMSC stemness markers, preventing their osteogenic adipogenic differentiation, maintaining high viability physiological cell-to-substrate dynamics. work presents proof concept data can be regulated living materials, using system based on symbiotic interaction different cells.
